Resumen: | La Furina es una proteína que se expresa en la mayoría de los vertebrados, su función es activar distintos precursores de proteínas. Trabajos previos han mostrado el rol crucial que juegan los distintos iones de sodio y calcio en la activación de la proteína Furina. Sin embargo, los aspectos energéticos y estructurales que determinan tal función han sido vagamente explorados. La comprensión a detalle de dichos aspectos podría resultar crucial para el diseño de nuevos inhibidores con potencial farmacológico. Este análisis no es sencillo de establecer debido a que es necesario desacoplar los distintos elementos energéticos. Para establecer dicho análisis se propone el uso de la teoría Perturbación de Energía Libre como herramienta que permita segmentar el rol de iones en el proceso de activación. A través de este análisis se encontraron las distintas propiedades energéticas de distintas conformaciones de la Furina, lo cual permite entender que componentes son importantes para la función y estabilidad de la proteína. Furin is a protein that is expressed in the majority of vertebrates, its function is to activate different protein precursors. Earlier research has shown the crucial role that the sodium and calcium Ions play in the activation of the Furin protein. Nevertheless, the energetic and structural aspects that determine its function have been barely studied. To fully comprehend this particular phenomenon might prove crucial in finding and developing new inhibitors with pharmaceutical potential. This is not an easy task; it requires decoupling the different energetic elements. To solve this problem, we propose the use of Free Energy Perturbations; this will allow us to find the role the ions play in the activation process. Through this analysis we found the energetic properties of different conformations of Furin, which helps to understand the importance for function and stability of different components of the protein. |
